The lluminati, an ancient secret society, anti-religious, resurfaces to complete his "historic destiny" - the destruction of the Catholic Church. They kill so cruel a famous scientist of an important research center to steal the first sample of antimatter ever conceived by man. The antimatter is one of the most coveted energy sources in the world but if not handled carefully, it becomes a substance with a devastating destructive power.

The findings symbols written on the bodies of dead scientists conduct research to Robert Langdon, a famous Harvard Professor of Symbology, secretly called to solve the mystery and discover the perpetrator of the crimes. Meanwhile, the capsule containing antimatter appears occasionally in closed-circuit television, hidden in an unknown location inside the Vatican City, causing a veritable pandemonium worldwide.
Also comes into play Vittoria Vetra, a mysterious and charming daughter of assassinated scientist, who will accompany Langdon to Rome to help the Swiss Guard to find the capsule. But things worsen further when they arrived, they discover that there is the election of the new Pope and the Illuminati have kidnapped four Cardinals favorites for the post.

They soon realize that the only way to deactivate the bomb is to find the perp and hope he does not kill the Cardinals before that. Therefore, Langdon and Vetra must solve a series of puzzles using the historical monuments of Rome, in hunting the invisible killer. Will they solve the riddles in time?
